Is it possible to get my 22-250 Ruger No.1 barrel reamed to 22-243 Middlestead?Will the barrel need to be backed off for the conversion?Can I just have the barrel reamed out with a 243 reamer and use it that way?Thanks in advance.

Ross Seyfried considered this about 15 years ago, and Ed (?) Shilen talked him out of it. It seems that the .243 case is only .133" (?) longer than the .22-250 case, and the .22-250 case walls are a little thinner, so by the time you make a .22-250 40-degree AI, there is virtually no gain to be had by the then popular version of the .22-.243. It could be that your goals will be best served by a .22-250 AI.
This is the limit of my expertise, so at this point I will yield to somebody who can discuss the relative volumes and performances of the .22-250, .22-250AI, .22-243, and .22-6mm with normal and heavier weight bullets. It's been suggested that there is little merit to driving normal weight varmint bullets much faster, and if you desire to throw heavier bullets, you begin to need to consider faster twists and longer barrels. Some varmint bullets will come apart at velocities just slightly higher than are normally achieved.

Yes you can ream your barrel to 22/243, and there will be enough "gain" to make it worth the effort. You cannot use a 243 reamer, because the neck diameter fir the 243 is TOO large.
